甲状腺癌组织端粒酶逆转录酶和P53基因表达及其临床意义

摘    要:目的 探讨端粒酶逆转录酶 (hTERT)和P5 3基因与甲状腺癌的发生、发展及预后的关系以及它们的相互关系。方法 应用免疫组织化学SP法检测 6 0例甲状腺癌、10例正常甲状腺组织、10例结节性甲状腺肿及 10例甲状腺腺瘤组织中hTERT和P5 3的表达。结果 hTERT和P5 3阳性率在甲状腺癌中增高 (P <0 .0 5 ) ;hTERT与P5 3阳性率在未分化癌、有淋巴结转移或Ⅲ、Ⅳ期病例中增高 (P <0 .0 5 ) ;hTERT表达与P5 3表达具有相关性 (P <0 .0 5 )。结论 hTERT与P5 3的过度表达在甲状腺癌的发生发展过程中具有重要作用 ,P5 3突变在转录水平上调控hTERT基因的表达 ,激活端粒酶可能是甲状腺癌发生发展的重要阶段 ;hTERT和P5 3在甲状腺癌中表达的变化可用于评估其生物学行为和判断预后。

Abstract:Objective To determine expression of h TE RT and P53 in thyroid carcinoma and its relationship with development and progno sis of the carcinoma. Methods A total of 90 samples of thyroid tissues (60 thyroi d carcinomas, 10 thyroid adenomas, 10 goitres and 10 normal thyroid tissues) wer e studied by SP immunohistochemical method. Results The positive-staining rates of hTERT and P53 were higher in thyroid carcinoma (P< 0.05) . The positive-staining rates of h TERT and P53 were higher in cases of undifferentiated carcinomas, positive metas tasis lymph nodes or at tumor stages Ⅲ and Ⅳ than in the cases of well-differ entiated, no metastasis lymph nodes or at tumor stages Ⅰ and Ⅱ (P< 0.05 ). The expression of hTERT was significantly related with that of P53 (P< 0 .05). Conclusion Overexpressed hTERT and P53 may play an important role in the carcinogenesis and progression of thyroid carcinoma and hTERT expression is related to P53 protein . Examination of expression of hTERT and P53 proteins may be helpful in judging the behavior and prognosis of thyroid cancer.
