摘    要:目的  1 磷酸鞘氨醇是新发现的细胞外递质和细胞内第二信使。研究它在人胃癌细胞与血管内皮细胞的增殖及凋亡调控中的作用。方法 光镜观察二甲基鞘氨醇作用前后的细胞形态 ;MTT法检测细胞的增殖能力 ;电镜观察细胞的超微结构及有无凋亡发生。结果 ①抑制 1 磷酸鞘氨醇生成后 ,胃癌细胞与血管内皮细胞形态发生明显变化。②二甲基鞘氨醇几乎可以完全抑制两种细胞的增殖。③一定剂量时可诱导两细胞凋亡。结论  1 磷酸鞘氨醇在人胃癌细胞与血管内皮细胞的形态、增殖和凋亡的调控中起重要作用。

Abstract:Objective To determine the role of Sphingosine-1-phosphate(SPP) in regulating the morphology,proliferation and apoptosis of human gastric carcinoma cell and human vascular endothelial cell. Methods The morphological changes were observed under light microscope.The proliferality of HGCC and HVEC was determined by MTT.The ultrostructural changes were observed under electron microscope.Results The morphology of both cells changed a lot after the synthesis of SPP was inhibited.When DMS was up to a certain dose, the proliferation of two cells could be almost completely inhibited and DMS could induce apoptosis of both cells.Conclusion Sphingosine-1-phosphate plays an important role in regulating the morphology,proliferation and apoptosis of human gastric carcinoma cell and human vascular endothelial cell.
