饮用水中过量氟对儿童唾液蛋白表达的影响

摘    要:目的 观察分别来自饮用水氟浓度为 3.6 8mg·L-1和 1.0 0mg·L-1两地区儿童间的刺激性颌 /舌下腺唾液中唾液蛋白是否存在差异。方法 测定受试对象的刺激性颌 /舌下腺唾液的唾液流率、唾液总蛋白浓度和单位时间唾液总蛋白的分泌量 ,分析唾液蛋白组成及各组分占总量的比例。结果 ①F3 .68组颌 /舌下腺唾液流率高于F1.0 0组 (P <0 .1) ;②F3 .68组颌 /舌下腺唾液总蛋白浓度明显低于F1.0 0 组 (P <0 .0 1) ;③F3 .68组颌 /舌下腺单位时间唾液总蛋白的分泌量显著低于F1.0 0 组 (P <0 .0 5 ) ;④在十二烷基硫酸钠 聚丙烯酰胺凝胶电泳 (SDS PAGE)不连续电泳条件下发现 ,两组颌 /舌下腺唾液共发现 10~ 12条蛋白带 ,其中B10 、B12 蛋白带的出现率在F3 .68组显著低于F1.0 0 组(P <0 .0 5 ) ,且B9蛋白带的量占总蛋白量的比例在F3 .68组显著高于F1.0 0 组 (P <0 .0 1)。结论 两组刺激性颌 /舌下腺唾液中有关唾液蛋白的指标存在量的差异 ,提示饮用水中过量氟对儿童唾液蛋白表达有影响

Abstract:Objective To investigate the difference in stimulated submandibular/sublingual salivary protein between two children groups who came from two districts where the fluoride concentrations in drinking water were 3.68?mg·L -1 (F 3.68 Group) and 1.00?mg·L -1 (F 1.00 Group). Methods We measured the flow rate, the concentration of total proteins and the protein volume secreted per min of stimulated parotid saliva, and analyzed the composition of salivary proteins and their compositional proportions. Results ① The flow rate of stimulated submandibular/sublingual saliva in F 3.68 Group were higher than that in F 1.00 Group ( P < 0.1 ). ② The total protein concentration of stimulated submandibular/sublingual saliva in F 3.68 Group was significantly lower than that in F 1.00 Group ( P < 0.01 ). ③ The protein volume secreted per min by submandibular/sublingual saliva in F 3.68 Group was markedly lower than that in F 1.00 Group( P < 0.05 ). ④ The submandibular/sublingual salivary protein patterns were different between the two groups. 10 to 12 protein bands were found. The incidence of B 10 , B 12 in F 3.68 Group was obviously lower than that in F 1.00 Group ( P < 0.05 ). The proportion of B 9 in total protein volume in F 3.68 Group was remarkably higher than that in F 1.00 Group ( P < 0.05 ). Conclusion The stimulated parotid salivary protein of the two children groups was different in volume.It indicates that the excessive fluoride in drinking water can affect children's salivary protein expression.
