脂质体介导c-myc反义寡核苷酸对人肝癌裸鼠移植瘤生长的影响

摘    要:目的 观察脂质体 (LR)介导c myc反义寡核苷酸 (ASODN)对高表达基因c myc的人肝癌细胞SMMC 772 1裸鼠移植瘤生长的影响。方法 人肝癌细胞株SMMC 772 1以 1.5× 10 6 分别接种于裸鼠背部皮下 ,第 14天均生长出瘤结节 大小为 (4± 1)mm]后 ,分为实验组、空白对照组和错配链寡核苷酸对照组三组。实验组用脂质体包裹的10 μmol·L- 1 浓度ASODN每天一次直接瘤体注射 ,连用 5d ,观察 2 8d。结果 测量裸鼠瘤体外表 ,发现第 5天ASODN组瘤体明显小于对照组 (P <0 .0 5 ) ,瘤质量在第 2 8天后ASODN组明显轻于对照组 (P <0 .0 5 )。组织学HE染色显示瘤体组织在实验组较对照组坏死明显 ,免疫组化结果显示C myc蛋白表达在实验组较对照组降低。 结论 推测应用脂质体包裹ASODN可能对人肝癌细胞SMMC 772 1成瘤的组织细胞生长有一定的抑制作用

Abstract:Objective To investigate the effect of LR mediated c-myc antisense oligodeoxynucleotides (ASODN) on the growth of human hepatoma cell SMMC-7721 in nude mice, in which were identified overexpression of c-myc gene. Methods After 1.5×10 6 human hepatoma cells SMMC-7721 were inoculated into each backside skin in 9 nude mice, all nude mice developed tumor node on day 14. Then we divided them into 3 groups: experiment, empty control and mismatch control. We injected directly 10?μmol·L -1 LR entrapped ASODN into the tumor body of the experiment group, MODN or water into control group for 5 days, and observed for 28 days. Results By measuring tumor body size, we found that the size of group treated with ASODN was significantly different from that of control at day 5 (P< 0.05). The weight of tumor between the two groups had significant difference (P< 0.05) on day 28. There were differences in histology by HE stains. Immunohistochemical detection showed that C-myc protein expression was decreased in experiment group. Conclusion LR entrapped ASODN may inhibit the growth of human hepatoma in nude mice by direct injection.
