精神分裂症患者ghrelin基因多态性与利培酮治疗前后患者体重和血脂变化的关系

目的观察利培酮治疗前后精神分裂症患者体重和血脂的变化及ghrelin(生长索)基因Leu72 Met(亮氨酸72蛋氨酸)多态性与体重和血脂变化的关系。方法对66例精神分裂症患者单用利培酮治疗10周,分别在治疗前和治疗后每2周测量身高、体重和腹围;在治疗前和治疗4、10周时检测血脂;PCR-RFLP(聚合酶链式反应-限制性片段长度多态性)技术检测ghrelin基因Leu72Met多态性。结果利培酮治疗期间,精神分裂症患者体重和血脂指标均有显著性增加(P<0.01)。体重和血脂指标的变化与ghrelin基因多态性无关(P>0.05)。结论精神分裂症患者利培酮治疗有体重和血脂水平的升高;Ghrelin基因Leu72 Met多态性和患者的体重和血脂升高无关。

Relation between the ghrelin gene Leu72Met polymorphism and changes of body weight, plasma lipid during risperidone treatment in schizophrenia

Objective To assess the effects of risperidone therapy on the changes of body weight and serum lipid in patients with schizophrenia or schizophrenia-like psychosis and to analyze the relation between ghrelin gene Leu72Met polymorphism and the change of body weight and serum lipid.Methods Totally 66 patients with schizophrenia or schizophrenia-like psychosis were recruited.Waistline and body mass index(BMI) were measured on admission and every 2 weeks subsequently;serum lipid was also measured on admission,4-week and 10-week treatment.The polymorphism of ghrelin gene was determined with PCR-RFLP technique.Results After 10 weeks' risperidone treatment,waistline,BMI and serum lipid increased significantly(P<0.01).There was not relationship between the changes of body weight and serum lipid and the ghrelin gene Leu72Met polymorphism(P>0.05).Conclusion After receiving risperidone treatment,patients have weight gain and increase in the level of serum lipid.Ghrelin gene Leu72Met polymorphism is not a risky factor of weight gain and serum lipid increase caused by risperidone.
