P~(53)5-8外显子在喉癌中的表达

采用改良玛尼亚法从喉癌患者术后新鲜组织标本中分别抽提全DNA，用PCR－SSCP方法进行扩增和电泳，然后行硝酸银染色，判断喉癌的中心组织、癌旁组织、转移淋巴结中的P535－8外显子的突变。结果表明：35例喉癌组织标本中P535－8外显子的突变率为60％，P53突变与淋巴结转移存在相关关系，P535－8外显子的突变与TNM分期及高中低分化均存在正相关关系，提示P53突变与喉癌的发生发展有密切关系。

INVESTIGATION OF MUTATION OF EXON 5-8 OF P~(53) GENE IN LARYNGEAL CANCER

We used improved Mania method toextract full DNA from laryngeal cancer tissue justafter operation. After culturing and gel electrophoresis by PCR-SSCP method, we stained itwith AgNOs. In this way, we could investigate thecentral tissue of laryngeal cancer, normal tissuearound the cancer and lymph node near it. The mutation of p53 5-8 exon also could be examed clearly.The results showed: mutation rate of P53 5-8 exonin 35 cases laryngeal cancer was 60%; mutation ofp53 was related to lymphatic metastasis ; mutation ofp53 5-8 exon was related to stages of TNM and differentiation. We conclude that mutation of P53 isclosely related to incidence and development of laryngeal cancer.
