丹参对大鼠腹腔感染状态下胃黏膜Na~+-K~+-ATPase和pH值的影响

摘    要:目的 探讨丹参注射液对严重腹腔感染状态下大鼠胃黏膜Na+ K+ ATPase活性变化和胃黏膜内pH的影响。方法 利用大鼠盲肠结扎穿孔造成腹腔严重感染动物模型 ,检测穿孔前和穿孔后 3、6、12、2 4、4 8h各时相大鼠胃黏膜Na+ K+ ATPase活性和胃黏膜内pH的变化。结果 盲肠穿孔后感染组 3hNa+ K+ ATPase活性显著降低 (P <0 .0 5 ) ,12h降至最低 (P <0 .0 1) ,仅为对照组的 4 8.5 % ,4 8h仍未恢复正常 ;丹参组 12h、2 4hNa+ K+ ATP酶活性比感染组显著升高 (P <0 .0 5 )。感染组胃黏膜内 pH值穿孔后 3h即有下降 ,6h明显低于对照组 (P <0 .0 5 ) ,12h下降至最低 (P <0 .0 1)。 4 8h仍低于对照组 (P <0 .0 5 )。丹参组从 12h开始胃黏膜内pH值与感染组显示出差别 (P <0 .0 5 )。结论 严重腹腔感染状态下胃黏膜Na+ K+ ATPase活性和 pHi降低可能是引起胃黏膜屏障功能受损的主要原因。早期应用丹参对有效地预防应激性溃疡的发生有重要的临床意义

Abstract:Objective To investigate the effect of Dansen on gastric mucosal Na +-K +-ATPase activity and pH during severe intraperitoneal infection in rats. Methods The intraperitoneal infection rat model was established by cecal ligation and puncture (CLP). Assay of Na +-K +-ATPase activity in gastric mucosal tissue was conducted by biochemistry method and the vitreous electrode was used to measure gastric mucosal pH. Results The activity of Na +-K +-ATPase was markedly decreased in the infected group at 3h after perforation, compared with the control group (P<0.05). There was a minimum of Na +-K +-ATPase activity at 12 h post-perforation in the infected group (P<0.01), only about 46.6% matched to the control group. At post-perforative 12 h and 24h , Na +-K +-ATPase activity of Dansen group significantly increased much more than that of infection group(P< 0.05). Gastric mucosal pH of the infected group decreased significantly compared with that of the control group (P<0.05) at 6h after perforation, and rapidly dropped to the lowest at 12h post-perforation(P<0.01). At 48h after perforation , pH was lower in infected group than that not only in the control group(P<0.05), but also in Dansen group(P<0.05).Conclusion The reduction of Na +-K +-ATPase activity and pH might play an important role in gastric mucosal barrier damage following severe abdominal infection induced by CLP. The application of Dansen in earlier period can prevent the occurrence of stress ulcer.
