硒对培养人肝细胞的毒性作用

摘    要:原代培养人胚肝细胞,观察外加不同剂量亚硒酸钠对肝细胞生长代谢的影响。结果表明,当培养液中硒的作用浓度增至1.156×10-6mol/L时,肝细胞聚缩、脱落,丙氨酸转氨酶外释增多,白蛋白合成减少,存活率下降。提示过量硒具有明显的肝细胞毒性作用。

Abstract:The experiment by culturing human fetal hepatocytes in the medium containing different level of sodium selenite was conducted to observe the effects of selenium on the growth and metabolism of hepatocytes. The results showed that over-does selenium (1. 156 ×10-6 mol/L ) could in-crease the alanine transaminase (ALT) activity in medium and decrease the survival rate of hepato-cytes and the albumin content in medium. Under the optical microscope, the morphylogical changes of hepatocytes caused by excessive selenium, such as disorder of cellular contraction and shedding, were observed. It is suggested that surplus selenium might cause the toxical change of hepatocytes.
