更年期大鼠Th1/Th2平衡与脾细胞凋亡

摘    要:目的了解更年期大鼠脾细胞凋亡与辅助性T细胞1(T-helper l,Th1)和辅助性T细胞2(T-helper 2,Th2)型细胞因子的增龄性改变。方法应用Annexin V-FITC/PI双染色流式细胞仪测定脾细胞凋亡率;ELISA法测血清γ-干扰素(interferon-γ,IFN-γ)、白介素-4(interleukin-4,IL-4)水平。结果更年期组大鼠脾细胞凋亡率(10.3%vs.7.7%,P<0.05)、血清IFN-γ质量浓度明显高于年轻组大鼠(23.80 ng/Lvs.10.92 ng/L,P<0.01);更年期组大鼠与年轻组相比血清IL-4质量浓度有下降趋势,但差别无显著意义(0.1296 ng/Lvs.0.3094 ng/L,P>0.05);更年期组血清IFN-γ/IL-4比值明显高于年轻组(183.64vs.35.29,P<0.01),Th1/Th2平衡朝Th1漂移。结论大鼠衰老过程中Th1/Th2平衡朝Th1漂移,脾细胞凋亡增加与IFN-γ水平增高有关。

Abstract:Objective To investigate the relation between the apoptosis in splenocyte and the changes of Th1/Th2 balance in serum of menopausal rats.Methods Twenty female SD rats were divided into two groups: menopausal group(n=10;15 months),young group(n=10;5.5 months).The apoptosis in splenocyte of rats was measured by Annexin V-FITC/PI flow cytometric assay.The levels of IFN-γ and IL-4 in serum were measured by ELISA.Results The percentages of apoptosis cells in splenocyte of menopausal rats were significantly higher than that of young rats.The serum IFN-γ level of menopausal rats was significantly higher than that of young rats(23.80ng/L vs.10.92ng/L,P<0.01).The level of IL-4 was similar in menopausal and young groups.Conclusion The increasing of apoptosis in splenocyte of menopausal rats is related to the increasing of IFN-γ levels with aging.
