中药安迪粉针剂对离体肿瘤细胞的细胞毒作用

摘    要:目的 观察以蟾酥和人参为主要配方的中药粉针剂安迪 (Andi)对肿瘤细胞的药物毒性作用。方法 采用MTT法测定Andi对 3株人肿瘤细胞的细胞生长曲线、细胞存活率 (SR)和细胞倍增时间 (TD) ;观察Andi对ECA 10 9细胞生长的时效关系。克隆计数法检测不同浓度Andi对ECA 10 9细胞在含氧和乏氧状态下的生存分数 (SF) ,计算其乏氧细胞毒性比 (HCR)和半数抑制浓度 (IC50 )。结果 ①Andi组和对照组对ECA 10 9、HepG2、SGC 790 1细胞的TD 分别为 118.72、12 3.0 8、4 0 .17h和 4 6 .12、4 0 .17、19.6 7h ;Andi组的SR分别为对照组的 33.33%、5 0 .5 4 %、36 .88%。② 12 .5 0、2 5 .0 0、5 0 .0 0 μg·mL- 1 Andi对ECA 10 9细胞在有氧和乏氧环境中的SF分别为 1.34、0 .97、0 .2 5和 0 .96、0 .2 7、0 .18。③Andi在乏氧环境中能抑制ECA 10 9细胞的生长 ,其作用大小与Andi浓度及作用时间呈正相关。④在有氧和乏氧下Andi的IC50 分别为 39.6 0 μg·mL- 1 和 2 3.5 0 μg·mL- 1 ,HCR为 2 .0 2。结论 Andi对多种人肿瘤细胞具有乏氧细胞毒作用 ,其程度与Andi的浓度和作用时间呈正相关

Abstract:Objective To observe the cytotoxicity of Andi on several human neoplasm cell lines. Methods Human esophageal carcinoma cell line (ECA 109), human hepatocarcinoma cell line (Hep G2), human gastric carcinoma cell line (SCG7901) were treated with culture medium with Andi (Andi group) or without Andi (control group) respectively in both air and 99.99% nitrogen. The cell survival curve, cell survival rate (SR), double time (T D) and relation of time-effect were examined by MTT assay. The cell survival fractions (SF) were tested with clone counting test on ECA 109 cell line. The hypoxic cytotoxicity ratio (HCR) and half inhibitive concentration (IC 50) were calculated. The effect of acting period of Andi on cells was observed from optic microscopy. Results ① In Andi group and control group, ECA 109, Hep G2, SCG 7901 cells' T D were 118.72?h, 123.08?h, 40.17?h and 46.12?h, 40.17?h and 19.67?h, respectively, and the SR in Andi group was 33.33%, 50.54% and 36.88% compared with control group. ② With cells of ECA 109, oxygenic SF and hypoxic SF detected by clone counting test were 1.00, 1.34, 0.97, 0.25 and 1.00, 0.96, 0.27, 0.18 in control group, Andi group with 12.5?μg·mL -1 ,25.0?μg·mL -1, 50.0?μg·mL -1 concentration respectively. ③ Andi was able to inhibit the growth of hypoxic ECA 109 cells and the effect was related with concentration of Andi (r= 0.984), acting period of Andi on cells. ④ The IC 50 was 39.6?μg·mL -1 to oxygenic cells and 23.5?μg·mL -1 to hypoxic cells. The HCR was 2.02. Conclusion With the condition and dosage in this study, Andi is provided with cytotoxicity on several human hypoxic neoplasm cells and the effectiveness has direct correlation with the concentration and the period of process of Andi.
